Yes you would setup the subdomain using cPanel first, then create the database using cPanel as normal.

Great - have done that now I want to access it but as the domain has not resolved and might not for a while, what would be the address I would go to to finish setting up WP?

I'd take a look at the Wp manual for that information.
If it were normally at domain.com/wp-admin/install.php then in your subdomain it should be at subdomain.domain.com/wp-admin/install.php
I don't know if that's the URL but I am going from memory. The docs should tell you where the installer is.

that's correct but I'm coming at it form an unresolved domain, so I have the numbered ip with the ~username. I'll submit a ticket so I can post the actual address.

You'll need to edit the config file to use your http://IP/~username/ if you are trying to look at it from an unresolved domain. If it is a subdomain then it would be http://IP/~username/subfolder/
where "subfolder is the web root of your subdomain.
